The data presented here is an extrapolation of a pre-specified analyses of eGFR slope from the DAPA-CKD clinical trial.
The decline in kidney function over time (eGFR slope) is on the causal pathway to kidney failure. Emerging evidence supports the eGFR slope as a viable surrogate for kidney failure in clinical trials of chronic kidney disease progression. Based on this, the aim of this prespecified analyses of the DAPA-CKD trial was to assess the effect of dapagliflozin on eGFR slope by establishing the effect of dapagliflozin on changes in eGFR during the acute phase (ie, from baseline to week 2 of treatment), chronic phase (ie, from week 2 to end of treatment), and total eGFR slope (from baseline to end of treatment full cohort). In the full cohort, dapagliflozin compared with placebo reduced the rate of eGFR decline from baseline to end of treatment by 0.95 mL/min per 1.73m2.
Extrapolation data are projections based on the eGFR from week 2 to end of treatment (DAPA-CKD chronic phase) and assume linear progression of eGFR decline. Initial eGFR decline over the first 2 weeks was estimated using eGFR slope data from the acute phase of the DAPA-CKD (baseline to week 2).
Example patient with initial eGFR of 30 mL/min/1.73m2 used eGFR slope data from the DAPA-CKD trial subgroup with eGFR <45 mL/min/1.73m2; eGFR at the start of the chronic phase: FORXIGA®, 27.4 mL/min/1.73m2; placebo, 29.6 mL/min/1.73m2. Example patient with initial eGFR of 60 mL/min/1.73m2 used eGFR slope data from the DAPA-CKD trial subgroup with eGFR ≥ 45 mL/min/1.73m2; eGFR at the start of the chronic phase: FORXIGA®, 56.3 mL/min/1.73m2; placebo, 59.0 mL/min/1.73m2.
